About Rapamycin Therapy

Rapamycin (sirolimus) is FDA-approved as an immunosuppressant for organ transplant. Its longevity profile rests on consistent lifespan extension across yeast, worms, flies, and mice, including in the NIA Interventions Testing Program. Human use for longevity is entirely off-label. Active monitoring of lipids, glucose, and immune function is essential. The PEARL trial (Sajjadi 2024) reported safety signals at 5-10 mg weekly in healthy adults but the longevity benefit in humans is not yet established.
